Light and Electron Microscopic Study of the Appendix in the Rabbit

Abstract
The appendix is a prominent example of gut associated lymohoepithelial tissue which is composed of non-lymphoid and lymphoid follicle associated portions. The purpose of the present study is to clarify the differences in morphology of epitelium between two regions by light and electron microscopy.
In light microscopy, the epithelium that lines the non-folicular region consisted of tall columnar cells and PAS positive goblet cells, which were similar to those of the mucosal epithelium of the large intestine. The boundary between epithelium and laminar propre was well marked and smooth. However, the epithelium of the follicular region showed low columnar or cuboidal cells, fewer goblet cells, and a marked infiltration of lymphocytes. The boundary between the epithelium and the underlying lymphoid follicle was irregular. The cells positive to acid phosphates and macrophages contained gram positive bacteria were always present in the lymphoid follicle but occasionally, in the epithelium.
In electron microscopy, the epithelium of the non-follicular region showed tall columnar absorptive cells and many goblet cells with abundant mucin granules. The enter-endocrine cells and a few lymphocytes were also found in the epithelium. The epithelium of the follicular region was composed of low columnar absorptive cells with many regular microvilli and numerous apical mitochondria, a few goblet cells, and follicle associated epithelial(FAE) cells. FAE cells, distinctive cell types in follicular region, were mainly found in intimate association of intraepithelial lymphocytes or macrophages, and were characterized by having a few irregular microvilli, many apical vesicles with or without dense contents, and some tonofilaments. An epithelial cell similar to FAE cell in general morphological feature was rarely found. This cell had less electron dense cytoplasmic matrix than that of FAE cell, and contained many tonofilaments (l0nm in diameter) in the supranuclear cytoplasm. Large number of lymphocytes invaded the follicular epithelium and came to lie in clusters mainly beneath the FAE cells. Main cellular components of lymphoid follicle were lymphocytes, macrophages, and plasma cells. Macrophages in this region often contained remnants of bacteria in the large vacuoles.
These results might suggest that the function of the epithelium in the non- . follicular region is similar to that of other intestine, but the function of the epithelium in the follicular region is mainly related to immune response.
